We will need more information. Starting with a schematic.

The 6NP3/2C51 is a different pinout from the 12AU7 and has about twice the gain of the 12AU7. When making this kind of change you need to articulate what you are trying to do. Is this simply for the sake of tube availability or are you attempting to get a specific response? We also need to know more about the circuit and what function the 6NP3 serves in it. Is it a small signal amp, a driver, a phase splitter, etc.? What configuration is it in? SE, SRPP, cascade, White follower, cathode follower, etc.?

All of these things affect the answer to your question. The best place to start would be posting a schematic. Even a simple hand drawn one would help. Can you do this?
